The news media is abuzz with stories about Alzheimer's disease clinical trials data and anticipated results. Let's peel back the layers of hype and expectation to explore why, more than three decades after the start of big investments in Alzheimer's research, there are still no safe and effective treatments that slow disease progression. The timing of key research findings, the official definition of Alzheimer's disease, the misinterpretation of human genetics, the continued use of an animal model without predictive validity, and potent cultural factors each played a role. What lessons can be learned that may be applicable to other sectors of biomedical science and where do we go from here? Finally, there are a few reasons for be optimistic, especially in terms of Alzheimer's disease prevention strategies.
